Ninomiya begon zijn carrière in de entertainmentindustrie toen hij in 1996 op 13-jarige leeftijd bij het Japanse talentenbureau Johnny & Associates kwam. Hij verliet het bureau pas in 2023. Voorafgaand aan zijn debuut als zanger bij Arashi in 1999, Ninomiya begon zijn acteercarrière toen hij werd gecast als Chris in het toneelstuk 'Stand by Me', dat was gebaseerd op de gelijknamige film. Sindsdien is hij verschenen in talloze televisiedrama-, film- en toneelproducties, waardoor hij bekend werd als de acteur van Arashi. Beschreven als een acteur die kan acteren met zijn mond en ogen, heeft Ninomiya een aantal prijzen en nominaties gewonnen voor zijn rollen.
In november 2019 trouwde hij met de voormalige omroeper Ito Ayako, twee jaar ouder dan hij. Hun eerste dochter werd geboren in maart 2021, hun tweede op 19 november 2022.
